Description
Bretazenil (Ro16-6028) is an imidazopyrrolobenzodiazepine anxiolytic drug of the benzodiazepine class. It was first synthesized in 1988 as a highly potent benzodiazepine that causes less addiction with a reduced profile of side effects, causes less strong development of tolerance. Structurally, bretazenil is close to the GABA antagonist flumazenil, although it has a different effect. It differs from traditional 1,4-benzodiazepines in that it is a partial agonist and binds to α1, α2, α3, α4, α5 and α6 subunits containing benzodiazepine GABA receptor complexes. 0.5 bretazenil is equivalent to 10 mg of diazepam, but has a stronger sedative effect.
Studies on primates have shown that bretazenil enhances the action of neurosteroids acting on the binding site of neurosteroids of the GABA receptor. Bretazenil causes a decrease in cortisol secretion and significantly increases the release of prolactin, has hypnotic properties, but worsens cognitive abilities, causes a decrease in the number of movements between sleep stages, at a dosage of 0.5 mg of bretazenil, REM sleep is reduced, and stage 2 of sleep is lengthened.
This drug has never been commercialized and is sold only as a research substance, a designer drug.
